How to Use Semax Nasal Spray (Dosage + Timing)

Semax is a heptapeptide, meaning it is too large to cross the blood-brain barrier effectively via systemic circulation. Oral delivery results in near-complete degradation by gut enzymes before any absorption occurs. So how does intranasal delivery work?
The olfactory epithelium — the tissue at the roof of the nasal cavity — provides a direct anatomical pathway to the brain via the olfactory nerve. Compounds applied to this tissue are taken up by olfactory neurons and transported to the olfactory bulb, then to adjacent limbic and cortical regions, bypassing the BBB entirely. This is the same route used by some viral pathogens (which is why this pathway exists biologically — it is not an artificial workaround).
Research using intranasal administration in rodents and humans shows significantly higher CNS bioavailability than subcutaneous injection for Semax. This is unusual — for most peptides, injection is superior. For Semax, nasal spray is genuinely the best delivery format, not merely a convenient alternative.

Prime the sprayer
If the spray is new or hasn't been used in several days, pump 2–3 times into a tissue to clear the nozzle and ensure consistent dose delivery on the first spray.
Tilt your head slightly forward
Do not tilt back. Tilting back sends the peptide down the throat and reduces nasal absorption. A slight forward tilt keeps the spray in contact with the olfactory mucosa.
Insert nozzle into one nostril, angling outward
Aim slightly toward the outer wall of the nostril (not straight up). This directs the spray toward the lateral nasal wall and olfactory epithelium, away from the septum.
Spray and gently inhale
Press the pump fully and take a gentle sniff — not a sharp forceful inhale. A gentle inhale draws the spray to the upper nasal cavity where absorption is highest.
Do not blow your nose immediately
Wait at least 5–10 minutes. Blowing your nose too soon physically removes the peptide before absorption can occur.
Repeat in other nostril
For bilateral dosing (e.g., 100mcg per nostril), repeat the process in the second nostril.
Best times
- • Morning, immediately upon waking
- • 30 minutes before cognitively demanding work
- • Second dose mid-morning if needed (before noon)
Avoid
- • After 2:00 pm (sleep disruption risk)
- • Evening or night use
- • Immediately before sleep (BDNF activation is activating)

- •Once reconstituted or opened, use within 4–6 weeks when kept refrigerated.
- •Protect from light — UV degrades peptide bonds. Store in original packaging or wrapped.
- •Never leave spray at room temperature for extended periods — peptides degrade faster above 15°C.
- •If spray separates or develops cloudiness/particles, discard.
