5-Amino-1MQ (5-amino-1-methylquinolinium) is a small molecule NNMT (nicotinamide N-methyltransferase) inhibitor developed through collaboration between research groups studying adipose tissue metabolism. NNMT was identified as an enzyme that becomes overexpressed in white adipose tissue during obesity, contributing to a 'metabolic sink' that reduces cellular NAD+ availability and impairs metabolism. Inhibiting NNMT with 5-Amino-1MQ was proposed as a way to restore normal adipose tissue metabolic activity.
NNMT methylates nicotinamide, effectively sequestering methyl groups and reducing NAD+ synthesis from this precursor in adipose tissue. 5-Amino-1MQ inhibits this enzyme, increasing NAD+ availability in fat cells, activating sirtuin pathways, and shifting white adipose tissue from a storage-dominant to a metabolism-dominant phenotype. It does not appear to have systemic sympathomimetic effects, distinguishing it from traditional stimulant-based fat burners.
