SNAP-8 (acetyl octapeptide-3) was developed as a cosmetic peptide alternative to botulinum toxin (Botox) for reducing expression lines. It is an octapeptide derived from the N-terminal end of SNAP-25, a SNARE (Soluble NSF Attachment REceptor) complex protein involved in neuromuscular signaling. By introducing a competing peptide fragment at the skin surface, it was proposed to modestly reduce the efficiency of acetylcholine release at neuromuscular junctions near the skin surface — producing subtle muscle relaxation without injection.
SNAP-8 competes with the endogenous SNAP-25 protein for integration into the SNARE complex at nerve-muscle synapses in facial muscles. Partial SNARE disruption reduces the completeness of acetylcholine vesicle fusion with the presynaptic membrane, modestly reducing the magnitude of muscle contraction in treated areas. The effect is far more subtle than botulinum toxin but is topically achievable — a meaningful distinction for cosmetic applications.

Clinical Cosmetic Studies
Studies with topical SNAP-8 formulations (at 10% concentration) have shown statistically significant reductions in wrinkle depth measured by profilometry over 28 days, with effects strongest in areas of high muscle expression activity. Reduction in wrinkle depth of approximately 50% in responders has been reported.
