DSIP (Delta Sleep-Inducing Peptide) is a neuropeptide originally isolated from the venous blood of sleeping rabbits by Marcel Monnier and colleagues at the University of Basel in 1977. The original hypothesis was that it was a specific 'sleep factor' responsible for initiating sleep. Subsequently, research revealed more complex roles including stress modulation, pain regulation, hormonal balance, and antioxidant effects. It occurs naturally in the human body and brain, with levels fluctuating in relation to the sleep-wake cycle.
DSIP has multiple proposed mechanisms: it modulates voltage-gated calcium channels, has opioid-like analgesic effects through DSIP-specific receptors, influences somatostatin signaling to modulate GH secretion, and appears to regulate the HPA (hypothalamic-pituitary-adrenal) axis stress response. The delta wave sleep induction for which it is named may reflect its modulation of GABAergic and serotonergic pathways involved in sleep architecture.

Insomnia and Sleep Architecture
Studies in subjects with chronic insomnia showed DSIP improving sleep onset latency, increasing slow-wave sleep percentage, and normalizing disrupted sleep patterns — effects consistent with both delta wave promotion and HPA axis modulation.
Withdrawal Support
Russian clinical studies explored DSIP in alcohol withdrawal, demonstrating reduced withdrawal severity scores and improved sleep quality during detoxification — consistent with its GABAergic and HPA-modulating effects.
Combined with Ipamorelin
Some practitioners combine DSIP with Ipamorelin at bedtime, hypothesizing that DSIP's somatostatin-modulating effect combines with Ipamorelin's GHRP activity for enhanced nighttime GH release and sleep quality.
