Nootropic

    Semax

Based on an ACTH fragment, Semax is used for cognitive enhancement, stroke recovery, and neuroprotection.

    Known Benefits
    • Focus and memory
    • Neuroprotection
    • BDNF increase
    History & Background

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) developed by researchers at Moscow State University as a derivative of the adrenocorticotropic hormone (ACTH) 4-10 sequence, which had been identified as the cognitively active portion of ACTH. Unlike ACTH, Semax has no adrenal activity — it was specifically engineered to isolate the nootropic and neuroprotective effects while eliminating hormonal side effects. It is approved in Russia and Ukraine as a nootropic and neuroprotective treatment.

    How It Works

Semax's primary characterized mechanism is the dramatic and rapid upregulation of BDNF and NGF (Nerve Growth Factor) in the hippocampus and cerebral cortex. These neurotrophic factors enhance synaptic plasticity, neuronal survival, and learning. It also modulates dopaminergic and serotonergic systems, which contributes to its nootropic and mood-enhancing effects. The nasal route delivers it directly to the olfactory nerves and from there to the brain, bypassing the blood-brain barrier.

    Research & Clinical Applications
    • Cognitive enhancement — focus, memory, learning speed
    • Stroke recovery and ischemic neuroprotection
    • ADHD management research
    • Optic nerve protection
    • Depression and anhedonia management
    • Neurodegeneration prevention
    Notable Research Outcomes

    Stroke Recovery Research

Russian clinical studies have demonstrated improved recovery outcomes in ischemic stroke patients treated with Semax compared to controls, attributed to its BDNF-elevating and neuroprotective properties. It is used clinically in Russia for this indication.

    Cognitive Performance in Healthy Subjects

Controlled studies in healthy volunteers show improved attention, processing speed, and short-term memory with Semax administration — effects consistent with elevated BDNF and enhanced dopaminergic tone.

    ADHD Alternative Research

Some integrative practitioners and researchers have explored Semax as a non-stimulant ADHD support, with anecdotal reports of improved attention and reduced distractibility without the cardiovascular side effects of traditional stimulants.

    Known Concerns & Considerations
    • Stimulating profile can cause insomnia if dosed too late in the day
    • Potential for dopaminergic sensitization with chronic use — tolerance patterns not well characterized
    • Not approved in Western countries — regulatory classification varies
    • Mood elevation effect can occasionally tip into irritability or overstimulation
    • Individuals with bipolar disorder or psychotic vulnerability should exercise significant caution
    Reported Side Effects
    • Mild irritation (nasal)
