Thymulin (also known as facteur thymique sérique, or FTS) is a nonapeptide thymic hormone identified in the 1970s by Mireille Dardenne and Jean-François Bach at the Institut Pasteur. It requires zinc for biological activity and plays a role in T-cell differentiation within the thymus. Thymic hormone levels decline significantly with age as the thymus involutes — a process completed by midlife in most individuals. Thymulin research fits within the broader framework of thymic restoration as a strategy for immune rejuvenation.
Thymulin binds to specific thymulin receptors on immature T-lymphocytes and promotes their differentiation into mature, functional T-cells. It requires a zinc ion as a cofactor for receptor binding activity. It modulates the balance between Th1 and Th2 immune responses, promotes tolerance mechanisms, and may have anti-inflammatory properties. Its activity is closely tied to systemic zinc status — zinc deficiency renders thymulin inactive.

Age-Related Immune Restoration
Studies in elderly subjects with low thymulin levels demonstrated partial restoration of T-cell responsiveness following thymulin supplementation, correlated with improvements in clinical measures of immune function. The effect requires adequate zinc status.
